Аннотация: Circulating tumor cells (CTCs) are rare cancer cells shed from primary tumors into the bloodstream and serve as valuable biomarkers for early diagnosis and monitoring of metastasis. Their isolation remains challenging due to extremely low abundance and interference from blood components. Here, we report a spiral-shaped microfluidicПоказать полностьюchip integrated with a chitosan (CS)-based biofilm functionalized with folic acid (FA) or a breast cancer–specific aptamer (BC-10) for selective CTC capture. The spiral-shaped design maximizes surface contact with blood flow while providing active binding sites for tumor cells and clusters. The device achieved ∼80% capture efficiency for tumor clusters with >95% cell viability. Pilot studies using blood samples from breast cancer patients demonstrated specific enrichment of CTCs compared with healthy donors, and capture efficiencies were consistent with flow cytometry results. This biofilm-integrated microfluidic platform offers a robust and customizable approach for CTC isolation, combining high sensitivity, biocompatibility, and potential for downstream clinical applications in cancer diagnostics and monitoring.
